Rui Pedro dos Santos Patrício was born on 15 February 1988 in Marrazes, Portugal. He grew up playing football in the youth academy of Sporting CP, one of Portugal's renowned clubs, and made his first-team debut at 18. Over the next twelve years, he became one of the most reliable goalkeepers in Portuguese football, playing 467 official matches for the club and winning five trophies, including two Portuguese Cups.

In the summer of 2018, Patrício made a notable move to English football, joining Wolverhampton Wanderers in the Premier League. His arrival matched the club's period of significant ambition after their recent promotion and investment. His play helped Wolves secure their Premier League status and compete in European competitions, earning him recognition as one of the top goalkeepers in England during that time.

In 2021, Patrício signed with AS Roma in Serie A under manager José Mourinho, also from Portugal. In his first season with the club, he was part of the squad that won the UEFA Europa Conference League in 2022, adding another European title to his achievements.
